WIA Bio-Manufacturing Standard Guide

A guide to the WIA-BIO-015 standard

Contents

Chapter 1The Bio-Manufacturing Landscape and WIA-BIO-015A map of how living cells are turned into medicines and other biological products, the handful of numbers that describe a production run, the regulatory ideas… Chapter 2Upstream Processing — Cell Lines, Media and Growth KineticsEverything downstream can only recover what the cells have made. This chapter builds the quantitative toolkit of upstream processing: choosing a clone, feeding… Chapter 3Bioreactor Operation and Critical Process ParametersHow a production vessel is configured, mixed, aerated, fed and controlled — and how each of those choices shows up in the WIA-BIO-015 data model. Chapter 4Downstream Processing and Quality AttributesFrom a turbid harvest to a concentrated, virus-safe drug substance: the unit operations of downstream processing, the equations WIA-BIO-015 uses to size them… Chapter 5The WIA-BIO-015 Data Model — Types, Units and SchemasEvery calculation in the earlier chapters ends up as a field in a JSON document. This chapter reads the standard's TypeScript type definitions line by line… Chapter 6SDK and CLI Interfaces and Calculation RulesWhat the reference SDK and CLI of WIA-BIO-015 actually compute, line by line: formulas, thresholds, placeholders, errors, and the places where the command line… Chapter 7Batch Operating Protocol and System IntegrationHow a WIA-BIO-015 batch moves through its states, how measurements become alarms and decisions, and where the standard's objects sit among the DCS, historian… Chapter 8Validation, Conformance Testing, Certification and What Comes NextHow a biologics process earns the right to be called validated, how a software implementation of WIA-BIO-015 earns the right to be called conformant, where…

About this book

A map of how living cells are turned into medicines and other biological products, the handful of numbers that describe a production run, the regulatory ideas that shape every decision, and an honest inventory of what the WIA-BIO-015 standard actually contains.

Before a reader can use a data standard for bio-manufacturing, it helps to know what is being described. A standard that encodes "cell density", "titer" or "Protein A step recovery" only makes sense to someone who knows why those quantities matter, where in the process they are measured and which decisions they drive. This first chapter therefore does two jobs. It lays out the physical process — from a frozen vial of cells to a filled container of purified product — and it maps the parts of WIA-BIO-015 onto that process, so that every later chapter can be read as a zoom into one region of the same picture.
